520	3		_aEn esta investigación se elaboró un manual operativo para el recuento de células somáticas en leche de vaca, por el método microscopia directa, la metodología para este manual fue basado en la NOM-F-700-COFOCALEC-2004 y la de Standard Methods for The Examination of Dairy Products, aplicable a toda leche cruda de vaca y a sus especificaciones de calidad. Se realizó en el laboratorio de lácteos del Departamento de Ingeniería Agroindustrial utilizando leche de animales del área de pastoreo del Departamento de Zootecnia, la muestra se tomo de el galón de recolección, en tubos de ensayo de previamente esterilizados y se llevaron al laboratorio en un lapso de 20 min; se prepararon las muestras rotuladas con el numero de vaca, en un baño de agua a 30-40 °C, se tomaron 10 µL de leche con la ayuda de una micropipeta y se depositaron sobre un portaobjetos que en la parte inferior fue colocada una placa guía de 20 mm X 5 mm con dos repeticiones, pasado el tiempo de secado se le añadió reactivo solución de Breed, por un tiempo de exposición de 15 min, pasado el tiempo se lavo y se seco el frotis para su análisis en el microscopio con un enfoque de 100 X, dependiendo el número de células contadas en las franjas se calculó con el factor de trabajo el número de células somáticas por cada mL de leche. Como resultado el recuento celular somático en los límites aceptables se asegura la calidad del producto final, mejores precios e incentivos para el productor, se incrementa el rendimiento en la elaboración de quesos y se alarga la vida de anaquel de los productos lácteos, se mejora la salud de las vacas y la rentabilidad de la ganadería.
